What is Triomune 40?
Buy Triomune 40 online, is a
combination of three drugs commonly used in the management of the human
immunodeficiency virus (HIV-1) infection. Both stavudine and lamivudine belong
to the nucleoside analog class of antiretroviral drugs. Both drugs act by
terminating the growth of the DNA chain and inhibiting the reverse
transcriptase of HIV
How does Buy Triomune 40 work?
Triomune
tablets contain a combination of 3 antiretroviral drugs stavudine, lamivudine
and nevirapine, which are effective only against RNA viruses (retroviruses)
like Human Immunodeficiency Virus (HIV). These three drugs each work by
inhibiting the action of the viral-specific enzyme reverse transcriptase (RT)
which is needed for the virus to be able to replicate. Retroviruses convert RNA
to DNA, which is then integrated into the DNA of infected immune cells so that
the virus can replicate. Nevirapine in Triomune tablets is a dipyridodiazepine
derivative non-nucleoside reverse transcriptase inhibitor (NNRTI); nucleosides
are the precursors to the building blocks of DNA and RNA. Nevirapine in
Triomune tablets acts by binding to a specific site on the RT enzyme, the
enzyme’s active site known as the NNRTI pocket, disrupting the catalytic site
of the enzyme, and blocking its action in a manner that is noncompetitive with
nucleosides. Stavudine and lamivudine are both nucleoside analogue reverse
transcriptase inhibitors (NRTI), which means that they block the action of RT
by competing with one of the nucleosides needed to transcribe the viral RNA
into DNA, becoming incorporated into the chain of newly synthesized viral DNA
and preventing it from growing (chain termination). Stavudine is an analogue of
thymidine and lamivudine is an analogue of cytidine. The combined action of the
three antiiretroviral drugs in Triomune tablets prevents the integration of the
virus into the DNA of the infected CD4+ immune cell by 2 different mechanisms,
which inhibits the replication of HIV so that no more virus can be produced and
this reduces the viral load (amount of virus). Triomune 30 slows down the
replication of HIV within your body but does not kill the virus already
incorporated in the CD4+ cells or prevent the spread of the infection to
others.
What are the side
effects of Triomune?
The most commonly reported side effects when taking
Triomune tablets are related to the individual drugs in the fixed dose
combination tablets and include:
·
Stavudine:
severe peripheral neuropathy, which is dose related and occurs more frequently
in patients with advanced HIV infection or who have previously experienced
peripheral neuropathy.
·
Lamivudine:
nausea, vomiting, headaches, abdominal pain and discomfort, peripheral
neuropathy, arthralgias, myalgias, skin rash, pancreatitis
·
Nevirapine:
development of rash and liver toxicity; both could be potentially life
threatening if severe and needs careful monitoring
·
A
more serious but less common side effect is a condition called lactic acidosis
which is excess lactic acid in the blood due
to liver damage, which may cause pancreatitis, liver failure, or renal failure

Pancreatitis

Dosing
Triomune is suitable for patients with HIV
infections. You should take 1 tablet of Triomune-30 twice daily, 12 hours
apart, if your body weight is less than 60kg. If your body weight is greater
than 60kg, you may need to take Triomune-40. You may only be prescribed this
medication if you have completed a 2-week lead-in treatment period comprised of
300mg of lamivudine daily, 30mg of stavudine twice daily, and 200mg of
nevirapine once daily. If you miss a dose, take it as soon as possible. Do not
double your dose.
Precautions
Following therapy using Triomune, you may experience certain health
complications.
Lactic acidosis: Marked by an
accumulation of lactic acid in the bloodstream, this condition is commonly
observed in women, obese individuals, and those with prolonged nucleoside
exposure. Fatal lactic acidosis has been reported in pregnant patients
prescribed with a combination of stavudine, didanosine, and other
antiretroviral agents.
Immune
reconstitution syndrome: This condition has been seen in some patients whose immune system
begins to recover and respond to previous opportunistic infections after taking
Triomune. This may result in overwhelming inflammatory responses.
Fat
redistribution: Patients on multiple antiretroviral therapies may experience the
accumulation and/or redistribution of fat. This can include central obesity,
the formation of buffalo humps, breast enlargement, facial wasting, and
peripheral wasting.
Pregnancy &
Lactation
Stavudine and lamivudine are classified under
category C in the FDA’s Pregnancy Categories, whilst nevirapine is classified
under category B. Therefore, pregnant women should only take this drug if its
potential benefits justify any potential risks to the fetus. This drug should
be used with caution when treating breastfeeding mothers.
Overdosage
Any suspected overdose should be immediately reported to and monitored by
a healthcare professional.
How should Triomune be taken?
How long should
you take Triomune?
You should continue to take your Triomune 30 for as
long as recommended by your doctor, and you will be monitored carefully to
ensure that you do not develop any serious side effects, in which case your
doctor will probably recommend that you stop taking your Triomune tablets.
Missed dose of
Triomune
If you miss a dose of Triomune tablets take it as
soon as you remember, unless it is time to take the next dose, then skip the
missed dose. Do not take a double dose.
